Proteinase K is a biocatalyst belonging to the serine protease family, named for its efficient degradation of keratin, which was initially discovered. First isolated from a saprophytic fungus in 1974, this enzyme exhibits extremely high proteolytic activity and broad substrate specificity, capable of degrading most proteins in their natural state, including keratin and collagen, which are difficult for other proteases to break down.

Unlike common proteases, proteinase K possesses exceptional environmental tolerance. It maintains high activity under a wide range of pH (4.0-12.5) and high temperature (50-70°C), and its activity is not inhibited by strong denaturing agents such as urea and sodium dodecyl sulfate (SDS), nor by ethylenediaminetetraacetic acid (EDTA); in fact, it exhibits even stronger activity in the presence of these reagents.
